is it possible to have the origin of a diagram at the bottom left and the y-axis increasing in value upwards?

Items' internal coordinates always increase top to bottom. You could still report them to your users in whatever other coordinate system needed. For example, set diagram.Bounds so it extends from negative Y at top side to 0 at bottom. Then show absolute-value Y coordinates for items in the UI, and apply negated Y if user enters coordinates somewhere.

It seems we also added a Ruler.NegatedY property at some point for scenario just like yours -
